Linker Types

Linkers connect warheads in PROTACs and payloads in ADCs. Choice of linker affects stability, cell penetration, and efficacy. Visual Linker supports property calculation and 3D visualization for common linker chemistries.

Flexible linkers

PEG, alkyl chains, and other flexible spacers increase conformational freedom. They can improve ternary complex formation in PROTACs but may reduce selectivity. Use Linker Properties to compare rotatable bonds (flexibility) and length.

Rigid linkers

Aromatic or piperazine-based linkers reduce flexibility and can improve proteolytic stability and potency. Molecular weight and logP from Linker Properties help compare rigid vs flexible options.

Cleavable linkers

Valine–citrulline, disulfide, and other cleavable linkers release payload in specific environments (e.g. lysosome, reducing). Cleavability is often described in literature; use Hydrophobicity Calculation and 3D Viewer to assess physicochemical fit.
